UCLA leads team that found key to Cushing, a life-threatening disorder
LOS ANGELES, May 31 () -- U.S. and Dutch researchers say they may have found a key to treating Cushing, a life-threatening disorder triggered by benign tumors in the pituitary glands.Study authors included Li Du, Marvin Bergsneider, Leili Mirsadraei, Steven H. Jonker of the University of Groningen in the Netherlands; and Michael Downes and Ronald M. Evans of Gene Expression Laboratory at the Salk Institute of Biological Studies in La Jolla, Calif.Cushing is a condition marked by progressive weight gain, excessive fatty tissue deposits and a rounding of facial features, known as "moon face." It can lead to diabetes, hypertension, osteoporosis, obesity and psychological disturbances.Cushing disease, more common in women than men, is associated with a three- to four-fold increase in the risk of premature death.The findings, published in the journal Proceedings of the National Academy of Sciences, found testicular orphan nuclear receptor 4 (TR4) was overexpressed in the tumors.The scientists discovered that by knocking down TR4 in mice, they were able to reverse tumor growth and excess adrenocorticotrophic hormone production, the study said.The findings could potentially lead to a drug therapy for Cushing, the study authors said.
